My boss wants me to travel about 200 miles with three 6 feet construction ladders on the top of my 2006 corolla. The ladders weigh about 25-35lbs each and I really think that it will mess my car up.

Don't be stupid. Don't put ladders on top of a car. Is very dangerous, and no rack is designed to put ladders on them, especially on cars. Like the first answer was, get a truck from your company you work for. Sounds like he's trying to make you look like a fool anyways.

if you have a rack you can probably carry 100-150 lbs safely. Don't do it without a rack, or make him sigh a contract that he will pay for any damaged to your car. Don't do it without a guarantee!

keep adding crap until the roof caves in
at this point you will know u have reached the weight limit
and can begin to angrily remove stuff
