How can she be sure that if she books and pays for the first room that there will another single room available? She is worried that if she does this she might not be able to book the second room.
Any suggestions?

If the website doesn't allow you to book two rooms in the same transaction, she would be better going to a travel agent. The independant agents are usually better with such things. She would save a lot of money if they were prepared to share a twin room.

talk to the travel agent and see what would be the best deal for them

i am not sure if i am allowed to give site details but i use cheap hotels.com and get good rates. try them
