From the GW get on the Major Deegan Expressway Northbound.AKA Interstate 87.Follow 87 north and cross
the Tappan Z bridge.Continue on until you reach Route 17
and 86. [only goes 1 direction] Follow 17/86 most of the way passing The Catskills region,Binghamton and Elmira.
Then continue on until you reach Interstate 390 towards Rochester.When you come to the New York Thruway
which is Interstate 90 go westbound.You are now about an hour and a half away from the falls.You want to pass the great city of Buffalo and pick up 190 North there will be signs directing you to the falls. I usually take the Niagara Falls Blvd exit. Route 62. There you will find many hotels and restaurant's of sorts.If you need additional info contact me I have been to the falls 4 times and plan on going back there soon.Have a great trip! It should take you less than 6.5 hours.

You can go by train. The ticket is cheaper if you purchase or reserve it several months before the trip. The problem is that there is no night trains to Niagara Falls, there are three trains that took between 8h 45 and 9 h 45 (the three trains have different trip times) to arrive there and leave NY at 7:15 AM, 10:45 AM and 1:45 PM. The bus is slower than the train, you must change of bus somewhere in two of the trips and the fare is higher! (US$ 78, train purchased with advance US$ 66). The advantage is that there is a direct night bus.

From New York, take the George Washington Bridge into Jersey, take your first right of the bridge onto Palisades Parkway, the take the exit NY State Thruway Interstate 287/87 towards Albany. Exit at Route 17 which is Woodbury Commons, go all the way to Binghamton, but don't go the route to Elmira like the other guy said, head north when you see the exit for Syracuse, once at Syracuse go west towards Niagara. The whole drive can be done in about 6.5 hours!
New York to Binghamton in 2.5, Binghamton to Syracuse in 1.5, Syracuse to Niagara in 2.5.
One stop for gas around Syracuse and you should be good.
I've done this drive but to Toronto and I can get from Toronto NYC in about 7 hours. Fastest time was 6.5 hours. You can't imagine how fast I went. Don't speed on Route 17 around Middletown and Goshen, the cops have nothing to do their but look for speeders. And be cautious speeding between Rochester and Buffalo. Of course my trips begin around 3am so I get to avoid all the cops.

If you have a car, drive. When you get to Niagara Falls, you look at the falls for fifteen minutes and you've seen it all. You can then spend some time visiting Canada. You may also want to visit Buffalo, although I cannot think of a reason why.
