I didn't find anything on American Airlines or Delta, but Czech Airlines apparently flies out of JFK in New York to Prague.
Typical flight time is about 8 hours +

BUT IF U DON'T LIVE NEAR THOSE CITIES THEN JUST GO FOR ANY CHEAP FARE AIRLINES.
I TOOK LOT (POLISH AIRLINE) TO CZECH B4.. CONNECTED IN KRAKOW.
