The watermarks, threads, and many other things. If you look at the face on a bill, you see the lines going around the face? That really says "United States of America" a bunch of times (I forget how many) but on a counterfeit bill, it would just all blend together to one line. On the new bills, colors are slightly different all across the bill, and there's also watermarks of the president's face on the sides. If you hold it up to the light, there are threads that say "$10", "$20", or whatever on them, and they're also at a different place on each increment of bills.
Look up a website for it, it's pretty interesting. David D's site is really good, but it's less accurate on older bills that are worn and faded a little.

Two simple test are.
A pen test, makes a brown mark on a real bill.
The other is so simple. You scrunch up the bill on an edge and rub it on white paper, if the green ink comes off, then it's real.
Also real bills have a thread through them and a water mark.

All bills have a metallic strip in the paper that you can see when held to the light. It has the monetary amount on it. (one on a dollar, five on a five dollar bill, etc.)

When I was working they gave us a certain marker and u put a line on the doller bill and if it showed up green it was fine and if it showed up black it was counterfit.

holograms and watermarks and at stores they have markers that write in different colors if it is counterfeit
