The biggest difference is in the size.

Cameras and other devices that could use the original MS Pro can use the Pro Duo with the Adapter.

However, some of the newest Sony cameras can only use the shorter Duo.

Some models of both the original Pro and the Pro Duo are labeled High Speed. These are capable of recording pictures faster, if the camera is capable of sending it faster.

Check your camera specs before spending extra money on a High Speed card. It might not be worth it.

There is no difference in media format. MS Pro is bigger and MS Pro Duo is smaller and requires and adapter to fit in a slot of MS Pro.
