I think you need to define "gun"
In world war 2 the largest naval gun was 18 inches and deployed by the Japanese
the US army today deploys a 155mm Howitzer
the largest caliber rifle is a .50 cal
The largest handgun is also a .50 cal
The largest gun ever constructed and put in use was 80cm gun named "Dora" during WW 2 was transported on by rail for the Germans
the largest gun ever attempted was rail gun by Iraq that would have had the ability to launch a shell and hit target hundreds of milles away

The biggest I've heard about was a 914mm mortar, "Little David", built by the US in the end of WW2, and meant to be used as siege weapon against the Japanese.
It was, however, never used.

The most powerful weapon is the Golden Gun, from 007 Goldeneye .Remember? Remember the game?
if you shoot somebody with the golden gun, they die instantly and it only takes 1 bullet.
