For a woman it just requires a physical examination of the genitalia and usually a "wet smear" which is where a sample of the vaginal discharge is placed on a microscope slide with a long Q-Tip sort of thing. Certain symptoms or "yes" answers to some questions asked by your physician might cause them to want to draw your blood (the only way to determine several STD's like HIV-AIDS)

The tests are different for females and males, at least the speciman collection any way. For females you lay on a doctors table and put your feet in sternums(sp), this will spread your legs open so that the doctor can view the vulva area, they do a visual check. Then they insert a speculum into the vagina to open it for speciman collection with a long cotton swab. They will collect from the uterine wall. Mind you dendending on the person there is from slight pressure to extreme pain for this. Those who experience the pain will know before hand..

A male simply stands in front of the doctor with his penis in view for the visual inspection, The doctor will then take a specimen sample from the inside of the penis with the same cotton swab.
Mind you the doctor has to insert the cotton swap to the penis for collection.
This is generally for syphillis, gonnorhea and HPV. For Hiv it is a blood test and will not be given unless asked for. I believe for herpes and chancre there is only a visual...Don't quote me on that last one though.

The doctor will have you get up on a table and put your feet in these stirups. He will then place a metal deal inside of you and open them up. He will tighten and position them so that he can stick cotton qtip swabs in there. He will also stick a couple of more things in there, like to swab you cervix and insides. They will also take blood test.

Then you all finish. It should be a really quick procedure. I advise to just relax, because getting those metal things put in you can be really akward and painful if you not relaxed.
