What is a 12 year old boy doing with a steady girlfriend? That is much too young. I didn't allow my boys to date until they were 16.
However, to answer your question, there is absolutely nothing wrong with cutting off his contact with her for a week or so, if that's what you choose. It's not going to scar him permanently.

At that age, they will break up the next day or week. It won't last! Give him something like no TV or no video games! No 12 year old really has a girlfriend or boyfriend!

First of all, any punishment rendered needs to fit the offense. Taking away something they value for a prescribed time period is a usual tactic, but it's difficult to take away a friendship (in theory) because you've shackled yourself with the burden of enforcement that is almost impossible to realize. And, unless she was somehow complicit in whatever he did to get into trouble in the first place, it may be unreasonably harsh. (Of course, even if it is too harsh one week isn't going to kill him, but you don't want the cure to any situation to be worse than the original problem. This penalty may cause him to act out even more.) Maybe a more equitable solution would be to suspend phone, TV, or computer privileges for the week instead.

At 12 he should not have a girlfriend.
At 12 he does not love her very much, he isn't mature enough to know what love is. He loves the way he feels when he is around her.
Punishment or no, he needs to spend time away from her and develop his own resources.
A lot of parents would think that just because they have this inappropriate relationship, they should not be allowed to see each other or talk on the phone. I'm not even saying that; I'm just saying not to let them spend so much time together.
And explain to them that at 12, they are not to hold hands, let alone cuddle or kiss. They are too young.

Sure is early fr him to have a girl friend...I don't think however that is a good punishment idea- it could turn him into a rebel!
instead, try something else like shifting his bedtime to an hour earlier than usual- i know that works!

No. It's a bad idea to use the girl as a pawn in punishing him. Ground him but keep his girlfriend out of the equation. Unless she caused the problems, using her as punishment is a horrible idea. Grounding him implies that he can't see her but by saying "Because you were bad, I am taking Jane away from you" is bringing her into your disagreement and that is not fair to the girl.

that was the only punishment that hurt my son. taking away something he loved for 1 or 2 weeks. usually though he would try to talk his way out of it. once i sent him to his room. he knew it was over until he learned to respect.. it does work. its hard as they grow older.
