Once a cat has urinated somewhere, it will often urinate in the same spot because it can smell the urine. Unless you clean the urine from the handbag completely, your cat is likely to urinate there again.
If you regularly carry your cat in your handbag, perhaps you just need to give the cat more opportunities to urinate in the litter box more frequently.

cats under stress often pee in places that smell familiar and safe. your cat will associate your scent with safety. you no doubt have carries your perfume in your bag at some point? a cats sense of smell is much more sensitive than ours. try keeping your handbag where your cat cant' get to it. pepper dust in your bag can work. if you still cant stop him there is a spray called "Feliway" that can be gotten from your vet which is effective about 99% of the time. have used it myself to stop our cat peeing in the back of the TV unit behind the TV set.
on the flip side of the coin your cat may be complaining that his litter tray is not clean enough. try using a wood pellet litter, it smells better, bio-degrades, is lighter in weight ( important in this age of paying for refuse by weight), and it disintegrates to powder on contact with liquid which makes it obvious when you need to clean his tray. if he doesnt like the timber pellet litter or you cant get it try using a shallow layer of his regular litter and changing it every day!!!! including washing the tray out with a de-odorising cleaner. something like detol surface cleaner or even a little lemon juice. dont use any cleaners with ammonia in them as the residual smell to a cat will smell like a soiled tray.

It is too late now with that particular handbag. The smell is there and he thinks it is where he goes. Disinfect the bag or throw it out.
Put a tray with newspaper down and keep your eye on him and when he wants to go put him straight on the tray, which you can move each day nearer to the door and then he will go outside.

Litter box is going to be the best bet. When all else fails, pee on someone else's handbag, discover what its like, figure out how you stop yourself from doing it, and apply the same technique on your kitten.

White pepper. It wont smell that bad for you but it'll stop the cat getting in. This worked when my kitten kept peeing in the corner.

don't let it go in your handbag, or just put it up higher so it can't reach. Maybe keep your handbag closed
