Basically, the movie is about an attack on the city of New York by some incredibly large creature as seen through the eyes of someone using a portable videocamera like you would see used at a party, etc.
It starts at a surprise going away party for some young guy and soon the fun starts as the crowd there reacts to what they think was an earthquake or some huge explosion. Soon enough though they realize that something far far worse is happening.
As for what the actual monster is or looks like is somethuing that is being hotly debated now because the movie opens next month (January 18th to be precise); you can find many websites discussing this right now if you try doing a search for 'cloverfield' (one site you can try for a look at the trailer is : http://cloverfield-movie-trailer.blogspot.com/2007/12/cloverfield-movie-trailer.html ).
