Penn State is a nickname for Pennsylvania State University in State College, although it's technically in the neighboring borough of University Park. Legend has it that Old Main is in the exact middle of the state (it's off by a few miles).
The University of Pennsylvania is about a century older and is located in Philadelphia. The two schools are not affiliated with one another in any way. The University of Pennsylvania is usually referred to by either of the nicknames of Penn or UPenn.
