Well, it depends on what you like -
If you are an Online FPS type of player then go for either Unreal or Orange Box
If you like third person shooters, and along with a good plot go for Uncharted
If you like a lot more your money and to laugh buy Orange Box
And you like a stealthy assassination game go for Assassins Creed
Personnally I would get Assassincs Creed, its just so original

From what you listed, the best PS3 game to get is Uncharted Drakes Fortune. I have Uncharted and it's a great game, plus it's a PS3 exclusive and you should definantly get it for that reason. The main story mode in Uncharted is pretty lengthly (10-12 hours depending on how much you explore), plus to unlock everything in the game you have to beat it on all of the harder difficulty modes as well as normal difficulty. The graphics in Uncharted are unbelievable, if you want to see what the PS3 is capable of, get Uncharted because you won't be disappointed.
I've heard a lot of people say that Assassins Creed get's pretty repetitive, the controls take a while to get used to, and the PS3 version is a little glitchy or something.
Unreal Tournament 3 looks good, but I would wait until the price drops on it to get it.
As for the Orange Box, I really don't like the way the game looks, but you may like it. The Orange Box comes with a few games.

ok i recently got a ps3 and really went through heavy documentation to decide what games were worth it...
uncharted drakes fortune is good, the problem is that i find it not very realistic, even if you shoot a pirate in the head he doesnt instantly die (how realistic is that)... assassins creed (i own it) i find extremely entertaining, the graphics are sweet the gameplay is nice, it can get a bit repetitive if you dont know how to play it but you will see that it is a very good game to play... i havent played unreal tournament 3 and the orange box so i wont express my opinion but il add 1 thing i hope can help you, Call Of Duty 4 Modern Warfare is seriously the best game out for ps3 yet (my opinion), it has fantastic non-online gameplay and the online gaming is just as good ... i think of it as an answer to halo... i suggest cod4 but it depends on your personal taste XD
