It is almost impossible to determine sex on a turtle until it is close to sexual maturity- for Red-ear Slider, for example, that is about 4 years old.
On sexually mature turtles, the male's belly shell (Plastron) will be incurved and his tail will be longer with the cloacal opening near the middle. Females have flat or outcurved plastrons with short broad tails with the cloacal opening near the base.
Some species have other indications.
Sliders and other 'pond' turtles- males have long foreclaws
Some box turtles- males have brightly colored eyes.
In most species, the males are brighter. In some species, the males are also bigger- but usually the females are the largest.

OK well no offense to anyone here but all the other answers, well i wouldnt follow them. First of all the male turtle will always have more of a concave ( under shell indention) than the female. Simply for mating purposes. Second the female will most likely have longer nails to dig nests. Third is that the males usually have dull eye colors such as dark browns or yellows. The females will have brighter eye shades such as yellow or red. Fourth is that males are bigger than females. Last is that the male will have a bright yellow or dark orange on their shells, females draker brown. Hope this helps, please list the type of turtle you have so i can give you more info on their type. What i have listed is for mostly all turtles especially eastern box, painters, and res. Good luck.
