I snaked out main line, all other drains are fine in house except that room.

"I'm not a Plumber", but get this often.

Is there a Clean Out Plug local to that bathroom plumbing, or a "P" or "S" trap? How CLEAR is the vent pipe that vents that bathroom plumbing?

You can rent a snake to unplug it. Your tub and toilet share a common drain.

I would remove the stool and check to see if something has been dropped into it, if it is partially closed it will cause that, you should also check the water level in the tank, insufficient water will cause it to not flush properly, if all else fails ROTO ROOTER.
