Lleyton Hewitt has long had the best topspin lob in men's tennis, and it's better than Roger Federer's. It's Hewitt's signature shot and he can hit it effectively off both wings almost every time. Patrick McEnroe and Jim Courier have both said that Hewitt's lob is the best in the business. Andy Murray is probably a good second though.
For the female players, there is no question that Martina Hingis has the most effective lob. It's a common practice for her to draw an opponent into the net with a dropshot only to lob right over them as soon as they get to it. With the retirement of Hingis, Jelena Jankovic probably has the best lob nowadays. She is one of the only players on the tour that can effectively lob 6'1" Venus Williams consistently.

For the men's side, Lleyton Hewitt is arguably the best at the topspin lob rather because he maintains more of an offensive than a defensive lob. Usually people use the lob to push a net person back into the baseline and give more time to recover for themselves; for Hewitt, he uses a deep and high lob rich with topspin to give it more of a drop. While other players can use lobs very well, Hewitt can pull one off more sufficiently than most players.
For the women, Justine Henin has a very well developed backhand topspin lob. Different from Hewitt (since he uses a two handed backhand), she uses a greatly disguised backhand lob. When she's about to perform a lob, it looks like any other backhand passing shot. Only when she drops her racket head into a low position does the opponent know that a lob is coming. Disguise and form are what give Henin one of the best lobs in the game.

Hewitt has one of the best lobs. For sure. But the best lob i´ve seen comes from David Nalbandian. I mean, just like Tsonga hits crazy halfvolley dropshots in every match, he hit lobs like other hit overheads. I´ve had the sparetime to research all the highlights available on youtube, i know, that doesn´t count errors and stuff. But i´ve seen a compilation with Hewitt lobs, and a lot of Hewitt matches, and ofcause many Nalbandian matches aswell. And i must say, Nalbandian hands down has the best lob. For women, i´m not so sure, i mostly care for mens tennis. Hewitt and Nalbandian i would say. Federer can ofcause produce magic lobs with his one handet backhand ect. But it´s not his bread and butter shot, like it sometimes is for Nalbandian when the opponent is at the net. Lastly i wanna point out, i don´t actually play tennis myself, because i can´t afford it, but i´m a serious tennis fan and love watching the game, so there you go.

Lleyton Hewitt on the mens, hands down. For a guy with a largely workman-like game, his lob is a thing of beauty. He rarely misses and he disguises the shot so well. Even the tallest players and best volleyers frequently fall victim to it.

I don't know why someone would mention Roger, as he rarely lobs and it isnt one of his best shots. On the mens side Rafael Nadal is very good but I would say the best is Andy Murray. On the womens side I think I Jelena Jankovic has the best lob. Both Jankovic and Murray employ this weapon often so if you have seen them play you know they have the best in the business.

justine henin-best 1 handed lob in the womens game, uses itto her advantage, deep and effective.
