Interesting question. It's like asking what fundamental bit of dogma, if shown to be false, would cause one's spiritual beliefs to crumble? I suppose if historians would conclusively conclude that the Jesus story is a myth it would be the jugular for many. As for me, I'm a Buddhist and the meditation practice works whether you believe in it or not. Zen Buddhists don't even teach about karma or reincarnation, so they aren't part of the jugular, either. And, of course, it works even if Buddha never existed. I suppose if they found out that our actions have no effect on our future it would dilute my motivation for Buddhist practice somewhat.
