Does he seem very withdrawn and depressed, or is he quite contented being on his own all the time? Some people are natural introverts, and need a lot of time by themselves. They get drained by too much company and activity, and really are happier alone, with a book or computer for company. If your brother seems basically fine, and is eating and sleeping well, and friendly enough to a few people when he meets them, then you don't need to worry.

On the other hand, if he used to be very outgoing, and seems to be a bit low, or isn't eating properly, then he may have some form of depression.

He doesn't want to "hang out" with your friends. If you can explain that you miss him and would like to do something with him and your friends aren't going to be around, then he might be willing to do something.

I know he's your brother, but a word of experience here about other guys who are like your brother. There are a lot of 21 year old guys who don't like to "hang out" all the time. They're usually the ones who start jobs that make the big bucks. You might want to pay a little attention to them. They sometimes don't have the best social skills, but they certainly become good husbands and fathers.

He's alright. All he has to do is pick up a newspaper or turn on the TV and with all the bad things that people are doing in the world, that should be enough to want to make anyone stay inside and keep to themselves.

It sounds as though he is depressed about something or maybe he enjoys staying glued to his computers or video games. Boys tend to do that.
You could try asking him if something is up, if not, just relaize that he is isolating himself for a while, it happens.

He's just a nerd, don't worry about it.
