I agree.. if only one choice, go to Asakusa. (which is pronounced "ah-sock-sa", not "ah-sah-ku-sa")
But only if you are a one-time tourist. Asakusa is really overrun with tourists these days and every vendor there is set up for the busloads.
It is cool to see the surrounding temple streets, etc but they are all selling little trinkets and crap. Still, we always ahve a good time there. My daughter loves to eat from the stalls, (Okonamiyaki is the BEST here!), and take a little "rickshaw" ride.. (i told you, they have really over-touristed it)
Second runner up would be Omatosando and harajuku area, (get off at either station), but only on a sunday, in good weather, when the Harajuku girls are out and everyone is strolling, shopping, eating at the cafes, etc. and then a nice walk through Meiji-jingu, possibly see a wedding party coming through... and lunch at hte BEST revolving sushi in Tokyo. (right there on Omatosando)

I'd go with the advice of two above for Asakusa. It has more of a traditional air about and the temple - Sensoji is one of Tokyo's best landmarks. There's lots of small market stalls in the area selling all kinds of souvenir items and food.

Asakusa Cannon Temple and the "temple streets" surrounding it. I go back there every time I'm in Tokyo.

Shibuya. Just to see the sheer masses of people crossing the main intersection infront of the station under the neon and video screens.
