It's a close walk about 10 minutes. Right outside of Penn Station there is a waiting line for cabs. There are so many cabs you won't have to wait. Yet 10 East 40th is a quick walk and you could see Bryant Park and the NY Public Library.

My friend, in this weather, its best you take the subway instead. From Penn Station, take the updown (northbound) #1, 2 or 3 trains (Red Line) to the Times Square-42nd Street.
At Times Square-42nd Street, go downstairs (follow the sign) and transfer to a Queens-bound #7 train (Purple Line) to 5th Avenue-Bryant Park.
Exit the station at the 5th Avenue end and walk south 2 block along 5th Avenue. 10 East 40th Street is right across the street from the New York Public Library-Main Branch.

Regardless of the weather, walk it. I took a cab once from 16 E 41st to Penn station and it took a half hour, the next time I walked it in about 15 minutes.
Traffic during the work day in this area can be hellish.

It's within walking distance (about 15-20 minutes), but in this weather you might want to take a cab. It get's really cold in the city as the wind whips between the buildings.
Yes, there is a line on 7th ave to get cabs - it's usually pretty long, but moves quickly.

By bus, you'll need the 34th Street Crosstown as far east as you can go. 450 East is at the FDR Drive. It's difficult to say how much time you should allow. The buses run very frequently at those hours (every few minutes), but traffic can be bad. Allow at least 1/2 hour, including waiting for the bus and walking to your final destination.

if you have the 15 minutes, then walk.
otherwise, if you really want/need to take a taxi, get one on the *8th* Avenue side, because 8th Avenue is already going *uptown*.
if you end up on 7th Avenue, just make sure you get a taxi *above* 32nd Street, because you'll need to go eastbound anyway before you go uptown. by the same token, you can cross 7th and try to hail a taxi already going east on 32nd. that may be your best bet in any case.
(btw, the M4 bus ends and starts on 32nd Street east of 7th Avenue, and goes uptown on Madison Avenue, but who knows how long that would take...)
any way you can cut the taxi's distance should save you time and money.
