To do it as closely as leonardo you will have to do quite a number of layers. I would suggest with your most delicate brushes to very lightly build the shadows with a slow drying medium. The lips do have a very lightly rose tone, i would suggest to apply the color in your next to last layer. Also everytime you do a layer you will have to wait a few days before applying the new one.

DA VINCI used a special type of painting style called sufmato and he he was not just painting with a brush he was giving those light touches through his fingers..........
