Clerks ask this to make sure that you found everything okay. If someone says that they couldn't find something I have either
a) if it's not busy...I retrieve the item myself and tell the customer where they can find the item for future reference.
b) I'll page another worker and ask them to retrieve the item
c) told customer the item was out of stock if that was the case or that we didn't carry that type of item.
d) sometimes items can be special-ordered and all you have to do is ask.
Cashiers ask this question because they are trained to or they just want to make sure you walk out happy. Good service often leads to repeat business.

I've had customers ask me where the milk was when I was working at a department store and it's located nearby the registers, we don't have a big selection of milk, to me I took it as if they were asking for assistance.

They will tell you where it is so you can go find it. They really aren't supposed to leave their stations. Push comes to shove, they might call a manager or make the bagger take you; but, they will not stop the line for you and you will have to go to the back.

If your'e standing at their counter, apparently you did. They have to ask after all. it's a polite thing to do. It's called customer service. If you couldn't find something, then yes, they would send someone to get it for you if you really wanted it.

yeah, they will probably either say its in this aisle and then continue ringing up your items while you go get it, or tell another clerk to go get you your milk =)

They might say, "Well for next time it's (here), if you'd like to go back and get some we can suspend your transaction."
