In theory yes you can, but you don't just simply apply for a work visa, you need a company to sponsor you and the required petitions need to filed and approved by the USCIS.
However, it is a violation of your status in the United States to actively perform or seek employment while entering the U.S. on a B-2 visa or the visa waiver program.

Yes, if you have a sponsoring US employer. Your employer can petition your H-1B visa, Form I-129 Petition for Nonimmigrant worker with the approved Labor Condition filed with the Department of Labor. You also need to file Form I-539 Application to Extend/Change of Nonimmigrant Status. It is a mulit step and long time consuming process and you may have to go back to home country before your Visitor visa expires. If you overstay, it will have huge negative impact on your worker's visa approval. Good luck!

Yes, if you already have a job lined up. but you then have to go through an interview with INS and believe me this is tough. The least little misdemeanor will be questioned and if you have been in serious trouble previously then you can forget it.
