If you played a Japanese PS2 game on an American PS3 it would not work.
PS2 games are region locked. This means you can't play a PS2 game that's from one region, on a PS2 or a PS3 that is from another region. So even if you tried to play it on a PS3, it would still be region locked.
PS3 games on the other hand are not region locked. So if you had a Japanese PS3 game and you tried to play it on your American PS3 it would work just fine.

PS2 games are still region locked on the PS3 even though PS3 games are region free. I'm not sure what would happen but you wouldn't be able to play it.
