There is not an assigned day that is going to be the cheapest. It is all a matter of supply and demand. Having said that, many business routes (i.e. Washington to New York) will have lower fares on weekends when businessmen are not occupying the seats.

It really depends on where you're flying. Most US destinations do not have Midweek/Weekend fares any more. You may get a lower fare on a weekend based on the lowest fare not being sold out, but the fares themselves are not any lower than a weekday unless they're running some sort of sale.

That said, vacation destinations typically offer lower fares if you travel Tuesday, Wednesday or Thursday. ie: Hawaii, Tahiti, etc.

Price does not change due to the day you are flying, it is whether or not the flight is full, whether the cheaper coach fares have already been sold, and how far in advance you book the ticket.
