Sorry, I was not clear. I have an existing 6" metal duct feeding my bedroom and I want to tap into the side of it to route a little air directly to my closet. What do I use to cut the 4" hole in the 6" duct? Is there then a transition piece I would use to connect the 4" flex duct to the hole in the 6" metal duct? Thanks for all the answers!

I have to agree with RICH on this one. I have examples I've made attempts in and went back and removed them.

The issue is simple. You reduce/diminish the efficiency to the bedroom; you add; even if slightly; more effort on whatever size your AC unit is.

Can you wall vent/register the closet? Can you do louvered doors?

You'll need to get a 6x6x4 'Y'. If they don't have one get a 6" 'Y' and a 4x6 reducer. Then disconnect and remove the 6" piping that goes into the bedroom vent, only take out one section about 4' long, between 2 taped seams. Connect the 'Y' to the metal pipe and run your 4" flex pipe to your new vent in the closet and some 6" flex pipe to your bedroom vent. There really isn't a good way to cut a hole in a round pipe and seal it, you'll loose more air than it's worth to try or all the air will go to the closet and not to the bedroom (Murphy's law). Splicing a 'Y' in will make the job easier and more professional.
Don't forget the insulation for the 'Y', the best way to wrap a 'Y' is to wrap it before you install it. Use the insulation from the piece of pipe you took out. Cut a big enough piece to cover the body, then cut a small hole for the 'Y' part out and wrap the 'Y'. If you cut a hole in the insulation rather than warp around the 'Y', you won't have to patch cover the 2" wide part between the 2 holes in the 'Y'.

You will need a 6 inch metal tee fitting and a 6inch x 4inch round reducer. This is probably not a good idea though. You will probably end up with an uncomfortable bedroom. For more info check out the ducts page at my source.

Go to your local Lowes store or hardware store and buy a T-fitting reducer connector for duct work. The top of the T is 6" and connector is a single 4" duct. This way you can connect it to your standard duct and reduce it to a 4" duct./

You'll need a 4" x 6" reducer. You can find it at home improvement stores it is metal , roughly bell shaped,and in the duct work department.

There is a transition piece in metal that will connect you from one to the other. Check with local heating and cooling people, if not, they will tell you where you can get it.
