The Downtown expansion pack (I believe it's called "Night Life", if my memory serves me correctly) is the only EP that allows your sims to drive cars. It also unlocks a downtown map as well.
For other Expansion Packs, you can only take a taxi to other community lots, on any map.
The University EP unlocks, well, a university, but it's only used for sims that either grow up from teens to "young teen", or new sims that you create to go to university.
The Open for Business EP has a shopping district that you can visit as well.
If I recall, the Pets, and Seasons EP's, along with all the "stuff packs" (such as Family Fun) do not add any new maps, but they do come with additional lots that you can add to existing maps.

just to add to TBG answer the new pack Bon Voyage adds 3 vacation spots(you cant drive to these area's you have to book a vacation and a shuttle picks you up) what it does do is allow you to do is walk to all and any community lots with the exception of the university you can walk from college to any lot . which is great if you don't have nightlife because there's no need to call a cab just click on Sim and choose walk to community or home.

TBG is pretty well spot-on. You must have Nightlife to have access to cars. You can drive downtown or to work, or you can take your children to school. If you also have Open for Business, you can travel to Bluewater Village (like traveling downtown) in your car rather than taking a taxi.
