Go with Uncharted Drakes Fortune for sure. I bought that game last Sunday and it's awesome. It's got stunning graphics, nice gameplay, and a good story to the whole game. It took me around 11 hours in total to beat the game on Normal Mode difficulty, so the game has a little length to it, and to unlock all of the secrets and unlockables in the game, you have to beat it on all difficulties.
As a matter of fact, I'm about to start playing Uncharted on Hard difficulty later on today. Uncharted Drakes Fortune is a must buy for any PS3 owner who likes action/adventure games.

Assassin's Creed is a great game. However, it's kind of slow-paced due to the fact that it's more of a stealth action game than a flat-out action title. If you don't mind that, definitely pick up Assassin's Creed.
Uncharted: Drake's Fortune is also very fun. It's more fast-paced than Assassin's Creed. There's much more action and it looks amazing. If you want a faster, more action oriented game, this is the game for you.
Folklore is a hit or miss. Personally, I played it and liked it a lot. I've heard people don't like it because it's also a slower moving game with not much action.
I've ordered these in the way I'd recommend them. Assassin's Creed should be your first pick-up if you don't mind the more stealthy type of gameplay.

I'd go with Assassin's Creed, I love that type of game. However, I'm also getting Uncharted: Drake's Fortune. I'd rent first just to make sure you like it before you spend $60 on it.

I just bought assassin's creed yesterday and it is absolutely amazing from the visuals, gameplay, and the story is so in depth.. it kind of reminds me of a strong mix of prince of persia and like a tenchu/metal gear type of game. All in all I had to try it for myself after reading so many mixed reviews about the game but I am real happy with my choice. Hopefully this helps you make your decision.
