I know what a prime suspect is, but I want to know how you become one. Is it when the majority of plausible evidence points toward one direction?

Can someone be called a prime suspect when you have only testimonies? and questionable assumptions based on knowing the person's lifestyle/personality?

Evidence. Like the gun has his/hers prints on it.. etc. stuff like that.

I think they call it "person of interest" now.
The wife/husband is killed you always check the other one for motive and opportunity.
If someone hears me threaten you, or I took out a insurance policy on you a week before your murder. All those things would make me the "prime " suspect.

Not so much evidence but relationship to the actual crime and lack of a good alibi. Motive and access land more people in prison than evidence. That is why there are so many innocent people in prison, motive, access and no alibi.

Yes! Also, if somebody says you did it, and they even have the WEAKEST POSSIBLE EVIDENCE against you (i.e. You were seen in the area), you'd be amazed to see how far up the list you go.
