The sound from a bass speaker is essentially non-directional, so it doesn't matter if the main speaker opening faces upward. If you are using a manufactured or properly designed speaker cabinet, the port can face in almost any direction.

If this is a do-it-yourself design, the key thing is to have sufficient separation of the port opening from the speaker opening behind the main panel, i.e., inside the enclosure. This is usually done with ducting, which can be made of partitions or piping. This forces the sound wave from the rear of the speaker to travel a minimum distance before it comes out through the port. If the ducting and port size are correct, this "rear wave" reinforces the one from the speaker front, and you get much better bass from the setup. (If the distance is too short, the waves can partially cancel each other out.)
