Actually, in this context, "measure" means "dance." Romeo has crashed a party at Juliet's house. He's watching her dance with another man. He's saying, "As soon as this dance is over (this measure is done), I'll watch to see where she's standing, then I'll go up to her and touch her hand, which will make me feel as if my hand has been blessed."

Romeo had just seen Juliet come into the party and was just awestruck by her. He described her in all sorts of metaphorical ways (she teaches the torches how to burn, a bright jewel, a dove among crows). "The measure done" means he's through with telling (or measuring) how beautiful she is. Then he says he will watch where she is standing and try to touch her hand. This will make his rough or clumsy (the meaning of rude) hand blessed by her touch.
