Do some research on the internet about puppies that are taken too early and show her all of the social and training issues that can come about from taking a puppy too early. If that doesn't work, then explain to her that whatever issues that puppy has is her fault and you will lose all respect for her if she gives up on that pup and rehomes it.

Not sure what state you are in but in most states if you buy or give away a dog or cat under 8 weeks old with out also getting the Mom you are breaking the law this is for the pups on health if it was a true breeder she would not let it go till it is at least 8 weeks just tell her for the health of the pup to leave it with the Mom this is when they learn what they need to know is from the Mom and litter mates

A puppy should be at least 8 weeks of age before it leaves its mother and littermates.
If the breeder your sister is buying from is letting pups go at 5 weeks, he's most likely a backyard breeder.
You can tell your sister that pups from backyard breeders usually come with more health problems and temperament problems than pups from responsible breeders; not to mention that it's been scientifically proven that puppies who leave their litter too early often end up with emotional and behavioural problems.

You've got some really good answers so far...with exception to those that think this is okay...
There are health risks and chance for some serious behavior issues (since proper social skills are learned all the way up to 12 weeks with mom and litter mates.) I will speak from experience that dogs with behavior issues takes A LOT of work, money, and time. And with her having such a large, muscular dog....the last thing she wants is a dog that lunges, snaps, growls, and tries to tear apart every other dog he sees. Or a dog that is so petrified it is difficult to even take him on a walk.
*As some others have mentioned, look into whether this is legal or not. Even if it is legal...you might want to make a report with the SPCA that the particular breeder is selling pups at that age.

Tell her that's not a good breeder. A good breeder would not let the pups go until they are at least 8 weeks old. At 5 weeks old they are just beginning to socialize and develop their personality and would make a much better dog if left with the litter until 8 weeks.

Tell her 5 weeks is dangerous for that puppy, it will not be completely weaned from it's mother and if it is they obviously pushed it to food to soon. Tell her wouldn't you rather let your dog get proper nutrients and the best start on life by staying a extra 2 weeks with it's mom, puppies also learn a lot of their social skills from momma dog that will affect it in life...doesn't sound like a very good breeder so I would be wary that the dog will be without health issues anyways. No decent dog breeder would let them go that early.

Tell her that it is important to the growth of the puppy. This is when the puppy learns socialization, pack mentality, and the dog's mother disciplines them in the way that only dogs can do. She will be doing her an injustice separating her too early and she will see the results years down line. The dog may develop behavior issues, confidence issues, etc.

definitely not a good breeder. any puppy of mine must be at least 8 weeks and been to the vet for check ups and shots and wormings. They need as much time with mom and siblings as possible for socialization. And also I prefer to let the mom wean them and know that they are eating solid food.

Look up the laws in your state - that is illegal in many states! And if it is illegal, report that 'breeder'! That sounds like someone who is just trying to make a quick buck!
Look up some articles online about the negative impact of removing a puppy from its mom/liter too early and give them to your sister to read.
I personally prefer 9-10 weeks to bring a puppy home.

The puppy should be no younger then 8 weeks old.
Most places it is illegal to sell a puppy under 8 weeks old.
Obviously this is a BAD breeder, they do not care about the health or the well-being of the puppies. They just want to get rid of them so that they are out of their hair. They want to get the money and that is all.
Let her know that puppies taken away too young are prone to behavior problems. They can have problems with fear and also problems with biting.
