Depends on the business really. I mean, if you have alot of tools or equipment that are sensitive to the weather, then you need a van. But if you are a carpenter and most of your tools will survive a little water go for the truck, get a tool chest to put in the back and keep your more sensitive stuff in there (like power tools)

A truck with a hard tonneau cover can be just as secure as a van but a van has more room inside and you can work inside it at times...depends on the trade, tools, weather,etc.. Fuel economy is pretty close. Vans can be easier to transport ladders or pipes.

Depends on what kind of work you do and how it is going to be used. If you are continually going to load and unload your equip, a truck would be easier. If you are going to store tools and supplies, then a van would be more secure. What does others in your trade use? That should give you your first clue.
