Everyone is going to say Magnolia. But I went there and my cupcake SUCKED. Don't know if I happened to get the only bad one in the bunch. But waiting in that LONG line in the summer for a small cupcake that wasn't very good. The gourmet grocery store Dean and Deluca has some good cupcakes. But they don't make them. They get them from another bakery. Go to citysearch.com and look up NYC cupcakes. There are some other GREAT places that specialize in cupcakes. One of them is owned by someone that used to work at Magnolia.

"best" is a matter of taste, i suppose, but i usually go to the west side branch of Buttercup, at 141 West 72nd Street, only because it's closest to me. Buttercup is apparently a descendant of Magnolia.
that address has an interesting history, because the old fashioned Eclair bakery opened there 1n 1939, and was still there into the mid 90's. then it became a Krispy Kreme until Buttercup moved in not too long ago
