It means a deficiency of some sort in that time of your life. For example: If your nails take 8 months to fully grow, and the spot is in the middle of the nail, then you had a deficiency about 4 months ago. If spot is at the bottom, then it was recent, at the top then it was closer to the 8 months

I agree with Tazzlove. It does not necessarily mean a deficiency of anything. Most likely it is related to an injury that you probably cannot remember. I would ignore it unless they become recurrent or on multiple fingers.

I don't think its a calcium deficiency, but rather an excess. It could also mean you injuried your thumb during the development of that specific location. It could also mean you were exposed to a toxin. Arsenic poisoning can result in discoloration of your nails. Exciting, I know, but hey, it happens. Don't quote me on this, but I think lead is that same way.

More likely you hit your thumb with something. Did you slam it in a drawer or something similar? Sometimes that causes the swelling to make an air pocket under the nail. It grows out as the nail grows.

I think it is to do with damage. I have broken most of my fingers and have them on the ones i have broken.
