I need to label the back of a hand made quilt and my fabric marking pen is dry, no time to buy a replacement. Will a sharpie work as well or will that fade more with repeated washings?

Test it first on a scrap of fabric and then wash it. Sharpies do have a tendency to "bleed" into fabric. You have created a quilt which is labour intensive. I would wait to do it properly and not risk ruining it.

Regular Sharpies fade, even without washing, after a time. If you have no other option, use it but keep the pen moving quickly so it can't bleed through and run.
Offer to retouch the labeling when you get a replacement fabric pen, if that's an option.
Is there anyone not too far away who'd let you use her fabric pen if you came for a very brief visit -- or who'd bring you one to use at a place you both have to go anyway? That might be an option, too.

If allowed to set, the Sharpie will work better than the fabric pen.
It takes a full 12 hours for the compound in a Sharpie to fully dry.
Most users disqualify, as they fail to place the Sharpie into untraviolet light before agitating the cloth. If this is the case, it ends up looking like you mixed an ink tanker and a t-shirt factory in a war zone.

Not sharpie pen but sharpie fabric pen. Sharpie makes a faric pen one that washes and one that dosent. GO pick one up.

Have you time to stitch your inititals into the back of it? A sharpie will work for a while but would fade away after 3 or 4 washes.

It might fade a little. Its a little hard to write on it with sharpie though. On fabrics i mean.

it works as well as a pen, but it messes up the sharpie later on.

Yeah, a Sharpie could posibly be even better. Those things are much more long lasting and user friendly than laundry markers!
