Grand Canyon -GCN *- is actually the closest It's about 7 miles from the S.Rim entrance station
Scenic Airlines [code HP] used to do the flight of las Vegas Now I think they're base out of Boulder City NV -nice people
Flagstaff FLG is the next closest at about 60 miles connecting service is out of
Phoenix -PHX which 150 miles S.of FLG
Las Vegas LAS is about 300 or so miles away
As is the case with everything there are trade offs
FLG & GCN will be much more expensive to fly into than either LAS or PHX
Car Rentals may be cheaper in LAS and so it goes
If you're going to be backpacking use Scenic
No point in paying for a car you're not going to be using
* GCN averages about 300 flights a day and it's all VFR

The Grand Canyon is a 5 hour drive from Vegas. Seems an airport at least in AZ would be the best choice.

Definitely Flagstaff. You can, however reasonably also fly into Las Vegas, Albuquerque, or Phoenix. These might be as much as 6 or 8 hours away, but the drive is fantastic.
