I would say the V-1 and V-2 rockets used by Nazi Germany during the Second World War. Many people died in accidents during testing, but more died due to the slave labor employed in the manufacture of the rockets. More concentration camp inmates died working in the Nordhausen rocket factory than the citizens of Great Britain did in the rocket attacks.

Can't remember the exact name, but it was what the Nazi's were developing during WW2. I remember that after further development of these rockets, they needed more labor, so prisoners from the concentration camps were used for more production. Luckily the prisoners started doing things like urinating on the electrical parts and removing screws to parts of the rockets. Reasons for all those videos showing us these rockets blowing up on the launch pad, or doing 180's and blowing up right where it was launched. Good luck. These rockets were eventually used against London, England.

Actually, while everyone thinks its the Atomic weapons of WW2, it actually isn't. There are several weapons you could be looking it, as this question presents you with the opportunity to show your researching merit. I would suggest two weapons:
1) the AK-47. Since its inception it has been used in every regional conflict around the globe.
2) Biological weapons. Long before bullets their were blood-soaked blankets dumped into city watertables. Biological warefare has been around for a very long time and has racked up a massive kill list.

I do not think any of the answers provided so far meet your description.
You call it a "weapon" which precludes tactics and vehicles
You did not stipulate "who" developed this weapon.
20,000 lives lost during development would pretty much preclude a modern American weapon.....It was probably developed by a country who used human test subjects. I am going to guess a CBR (Chemical, Biological , Radiological )
weapon.....developed by America and used on the American Indians.......Smallpox

NOTE: While the Atomic bomb (nuke or Nuclear) did not kill anyone during development that I know of.....when it was actually employed it kill far more than a mere 5,000 ( 159,000 in Heroshima alone)

none. the bomb the Germans were developing before WWII killed thousands of slaves before the finally used it. Only because the had to build a shelter to make it and they dug out a mountain-shelter for its housing.

or It may have been the V2
