It would help if you could be more specific, as there is so much to see in Lisbon. Here's a list of things you can see there.
You can visit 'Parque das Nacoes', which is the former Expo 98, you can spend the whole day there, and if you get tired you can always go to Vasco da Gama shopping Mall or visit 'Vasco da Gama Tower'.

Accessibility to this Park is great as you have 'Gare do Oriente' right next to it and this station is worth visiting, because it was made by the famous Spanish architect Santiago Calatrava.

You can go to 'Belem' and visit Jeronimos Monastery, 'Padrao dos Descobrimentos', 'Belem Tower'

'Sao Jorge's Castle',
http://pt.wikipedia.org/wiki/Imagem:Castelo_Sao_Jorge_Lisboa_2.JPG
'Bairro de Alfama' where the traditional 'fado' was born, so I recommend you to go to one of their bars and listen to 'fado'
http://pt.wikipedia.org/wiki/Imagem:Alfama-CCBY.jpg
You can also visit the Cathedral

Go down to Rossio and visit 'Carmo's Convent'
http://pt.wikipedia.org/wiki/Imagem:Lisbon09.jpg
Go to Alcantara and visit the 'Aqueduto'

http://pt.wikipedia.org/wiki/Imagem:Aqueduto_das_%C3%A1guas_livres.JPG
You can also go for a trip in the 'Tejo' (Tagus) river.
You can visit the 'National Pantheon' where our most famous 'fado' singer - Amalia Rodrigues - is buried.

This Pantheon also has the name of 'Santa Engracia church', and we have a very funny saying about it. Check the link below for its history.

You can visit 'Chiado' a street that burnt down in 1988 and was rebuilt again, it is Siza Vieira's project (the most famous architect in Portugal)

There is so much to do and visit in Lisbon. And if you are interested in going out of town, I advise you to visit 'Cascais' and 'Sintra'.
Regarding food, you MUST eat some 'pasteis de Belem' in Belem, they are unique in the world.
If you want to visit only one place, then I would short-list to ''Parque das Nacoes' (because it is recent, and quite popular) and 'Belem' (because it is historical, next to 'Belem Palace' where the President lives and you can always stuff yourself with 'pasteis de Belem' ;-D)

Lisbon is lovely! First thing, get yourself a DK Eyewitness guide, it's the best and most informative guide book available, and comes in different languages.

We stayed in Pensao Del Mar in the Alfama district, and it was lovely. In the ground floor of the building is a cafe that we went to for breakfast every day except Sunday. Just near this Pensao is a lovely restaurant on the corner that serves delicious home-cooked Portuguese meals, including the one that I loved: pork cooked with clams. It was to die for! The seafood in general was delicious.

The Alfama district is great, it's all steep narrow streets winding through houses, very few places are accessible to cars.

There is one tram that takes in a lot of the city, and I can't remember the number offhand, it's in that guidebook in any case. (MIND YOUR BELONGINGS!!!! In most cities, the public transport system is used by pickpockets.)

I really liked Lisbon, I found the people so friendly and most people had enough English to make it easier to communicate.

For your information, we found that accommodation through www.hostelworld.com.
