He wants to wipe Israel off the map, and he has stated this on numerous occasions. I watched him being interviewed a few weeks ago by a British journalist, and again the Iranian despot declared that Israel 'must vanish'.
Israel is indeed the only nation with the courage and sheer chutzpah to do anything. The world should be glad that she had the guts to take out Saddam Hussein's nuclear facility back in the 80s.
Israel also has the most to lose if Iran gets nuclear materials, because she is top of their hit list. And as Iran supports and funds terrorist group Hezbollah, if the material falls into their hands then it is innocent Israelis who would suffer, G-d forbid that it should ever happen.

Iran is a terrorist state that has proclaimed that it wishes to destroy Israel. Nuclear weapons would give them the opportunity to do so, heaven forfend.
If Israel does indeed have a nuclear capability, it will use it only as a last resort, after a first strike by some other power.
Iran is the biggest threat to the West today, and their military capability must be curbed by the West.
Thus, the United States' interest is identical to that of Israel and of the entire free world.
In addition, individual terrorists must be eliminated--if necessary by engaging them and killing them.
There can be no rational argument against this two-pronged approach.
