The R group is the differentiating areas of the amino acid. Everything is the same except the R group.

To add to what ATP man said, the R group not only identifies which amino acid you have, but it also is responsible for the final protein shape (tertiary structure) of a single amino acid chain as the R groups form bonds with each other.

Since the function of proteins (especially enzymes with the "lock and key" specificity of binding sites) is a function of their shape, having the R-groups of the proper type and in proper sequence is important to maintaining the proper function.
