My experience has been mainly with ISP using JTAG. The STK300 supports ISP so pins used for JTAG are

Vcc
Gnd
Reset
CLK
MISO
MOSI

Check the data sheet for PINout diagram . Data sheet also details how to program using High Voltage programming which I assum the socket of the STK300 can perform.
