It means that someone has added something to the drink - usually alcohol, but it can also be a drug (such as GHB, or the "Date Rape" drug).
The term applies to both a known addititive (when EVERYONE knows that there is vodka in the punch - "The punch has been spiked") as well as the (ILLEGAL) unknown additive...

A spiked drink is when someone slips alcohol in it when they are not suppose to. For example, a highschool prom. If you get a class of punch and slip some vodka in it.....that is spiking the drink. Another "spiked" drink is when someone slips something into your drink, like the date rape drug.

It is a drink that has alcohol mixed into it, that would normally not be expected to have alcohol in. Like for example a punch at a high school party. There is supposed to be no alcohol, but if it is spiked, it has alcohol.
