No. There is no skin breakage like with girls or way to "test" sperm to see if they have had sex yet. Usually young men at the beginning of puberty have ejaculated sperm and semen more than once because of masturbation or nocturnal emissions(wet dreams). The difference with men is when they have sex (as different to masturbation) is they are "getting off" by putting the penis inside of another person instead of doing it by themselves. They are just breaking the personal, emotional, and sometimes moral barrier by having sex.
The only way to physically tell a male has had sexual contact, is if he have "left over" DNA evidence to test after wards, should the girl hand herself over to the proper authorities right after it happened and a rape test kit was used on her.
Now, should someone have their sperm COUNT tested at a clinic, the only thing a low sperm count would prove is that they may masturbate alot, or just masturbated hours before the test sample taken. Or he may possibly have a genetically low sperm count.

There is no way to know if a guy has had intercourse yet. Keep in mind that most guys like to severely over exaggerate the # of partners they've had.
