Well you need to start with a clean just open water bottle as a constant. Swab the rim and then rub the swab on a petri dish. They make petri dishes with bacteria food the helps the bacteria to grow. Then drink from a waterbottle, wash it like you would if you were reusing a water bottle, and do another swab on a petri dish. Check in a couple days to see if any bacteria grows.
