Yes he loses the ability to understand Parseltongue, J.K Rowling mentioned this in an interview, where fans had the opportunity to ask her questions. She also talked about what became, of some of the character's who's fate's weren't revealed in the epilogue.
"Nigel: Can Harry speak Parseltongue when he is no longer a horcrux?
J.K. Rowling: No, he loses the ability, and is very glad to do so."

According to an interview, JK confirmed that Harry did lost the ability to speak Parseltongue because the connection between him and Voldemort was already destroyed.

Actually after Voldemort had "killed" Harry, he killed the part of his soul that lie within him; with that bit of soul being gone, Harry no longer can speak parsletongue nor will he be having any more voyages into Voldemorts mind. Now the very last of Voldemort's soul was in him, the part that could not be put into a horcrux. Voldemort died simply because the elder wand refused to defend him.

While he was not the last horcrux destroyed, Jo said yes, he loses those abilities gained from Riddle's soul. No more talking to snakes.

Yes it does. And in some interview Jo confirmed this and said Harry is quite content not to be able to speak to snakes anymore. If you want to find it, I'd look on mugglenet.com or the-leaky-cauldron.org

Well it looks like your question's already answered. Yes, it's true . Without Voldemort trying to kill him when he was a baby, Harry would have never been able to talk to snakes in the first place. and now Voldemort's gone, so...... you do the math

Yes, that was mentioned somewhere in one of the chats/interviews that JKR has had since DH came out.
