It can and should be carried out both one eye at a time and binocularly. Although it is rare, some people have colour vision in one eye and colour vision in the other. If it is done with only one eye, the colour vision in one eye can mask the colour blindness in the other eye.

Colur blindness that has a genetic origin is probably always binocular. It is unlikely that a person has colour blindness in only one eye.
In any case one can take the test viewing the test cards through one eye at a time.
