Your fans may need cleaning (be extra careful when doing this) and you may want to delete some extraneous stuff off of your hard drive so that your computer doesn't have to work so hard. You should check to see how many programs are starting up when you turn on your computer and only limit the computer to what you really need at the time of start up. I would first check the fans and make sure your computer base is in a well ventalated area.

You probably need to run disk defragmenting (Start /All Programs/Accessories/ System / Disk Defragment ) The files that are on the disk are broken into small pieces that are scattered all over, so the drive is having to jump the heads a lot to go from section to section. Defrag will get all the pieces lined up in order. The first time in particular, it may take several hours to get the job done.
Delete all the files you can and empty the Recycle Bin before starting defrag.

Have you done a disk defragment? If not click on start, all programs, accessories, disk defragmenter. I don't know if doing this will help stop the noise but I do know it will help your PC. I have a 500GB external hard drive that has very little on it but it still makes noise sometimes, the noise may just be the fan in your PC so I wouldn't worry at this stage ;-)

I think you need to unplug it and take the side off of it and use air duster to blow all the dust off the fans 1 year of dust causes a lot of noise. Also clean the power supply fan.

When you clean your fan hold the fan blades still while you are using the air duster. If the fan blades spin the wrong direction you could damage the engine.
