Probably your teacher is wanting "Bacteria have no nucleus". But there are so many differences that it's hard to say. I'd guess that's the one though.

Bacterial cell has no nucleus, and the eukaryotic cell does. There are a bunch of differences, but this is the main one.

2. Bacterial cell has its DNA free-floating in the cytoplasm as rings of DNA called plasmids.
3. Bacteria have no membrane-bound organelles like mitochondria.
4. Bacterial cell wall is made of proteoglycan (animal cells have no cell wall; plant cell wall is made of cellulose).

Bacteria have no nucleus and no organelles with membranes around them (no mitochondria, no lysosomes, no endoplasmic reticulum, no chloroplasts, no Golgi bodies, etc.)
Eukaryotic cells have the nuclear material enclosed in a membrane, and eukaryotic cells have organelles bound by membranes.

prokaryotes have NO nucleus, NO membrane-bound organelles. Or if you want to go more in depth.. bacterial cell walls contain PEPTIDYLGLYCAN which consists of NAG and NAM (abbreviation)

WTF? There's hundreds of differences, and I'd be hard-pressed to characterize any of them as the "main" one. Yes, bacteria have no nucleus, but one could as well point to the fact that they don't have that they don't have mitochondria, golgi, endoplasmic reticula, or any other organelle as just as significant. Or the fact that they have circular chromosomes rather than linear chromosomes.
I find the question's reductive simplicity insulting to the student.
