Two choices - Boat or Plane

Spirit of Tasmania - operates between Melbourne and Devonport, sailing from both locations most nights and some days during peak periods, http://www.spiritoftasmania.com.au

Plane - http://www.qantas.com.au or http://www.jetstar.com.au

How much? Well that depends on flying, what city you fly into from Melbourne, what time of the day, week etc... as we know, no airfare is the same. Same goes for ferry, there's always off peak, during peak and hot deals...

The Spirit of Tasmania mate.
Its great, around 12 hour trip, you can purchase a sit up seat or take a cabin /and/or take your car too.

Other alternative is fly from Tullamarine or any of the other smaller fields, Virgin, Qantas, Jetstar and soon Tiger fly into Launceston (45 min trip approx). Or fly into Wynyard or Devonport or of course Hobart, take your pick, some good deals out there. Tassie is absolutely beautiful and worth the trip. Ive lost track of the number of trips Ive made there.
